چکیده
Previous attempts to establish transgenic mouse models to study the functions of transforming growth factor b1 (TGFb1) in the skin revealed controversial roles for TGFb1 in epidermal growth (inhibition vs. stimulation) and resulted in neonatal lethality in one instance. To establish a viable transgenic model for studying functions of TGFb1 in the skin, we have now developed transgenic mice, which allow focal induction of the TGFb1 transgene in the epidermis at different expression levels and at different developmental stages. This system, termed ‘‘gene-switch,’’ consists of two transgenic lines. The mouse loricrin vector targets the GLVPc transactivator (a fusion molecule of the truncated progesterone receptor and the GAL4 DNA binding domain), and a thymidine kinase promoter drives the TGFb1 target gene with GAL4 binding sites upstream of the promoter. These two transgenic lines were mated to generate bigenic mice, and TGFb1 transgene expression was controlled by topical application of an antiprogestin. On epidermal-specific induction of the TGFb1 transgene, the BrdUrd labeling index in the transgenic epidermis decreased 6-fold compared with controls. Induction of the TGFb1 transgene expression also caused epidermal resistance to phorbol 12-myristate 13acetate-induced hyperplasia, with a reduction in both epidermal thickness and BrdUrd labeling compared with those in controls. In addition, TGFb1 transgene expression induced an increase in angiogenesis in the dermis. Given that the TGFb1 transgene can affect both the epidermis and dermis, this transgenic model will provide a useful tool for studying roles of TGFb1 in wound-healing and skin carcinogenesis in the

I-43: Identification of SOX3 as an XX MaleSex Reversal Gene in Mice and Jumans
Background: Mammals utilise an XX/XY system of sex determination in which the Y-linked gene SRY (Sexdetermining region Y) exerts a dominant masculinising influence on sexual development. Sex chromosome homology and comparative sequence studies suggest that SRY evolved from the related SOX3 gene on the X chromosome, although there is no direct functional evidence to support this hypothesis. The ...

Transforming growth factor-b1 (TGF-b1) is expressed in the adult and embryonic vasculature; however, the biological consequences of increased vascular TGF-b1 expression remain controversial. To establish an experimental setting for investigating the role of increased TGF-b1 in vascular development and disease, we generated transgenic mice in which a cDNA encoding a constitutively active form of...

To assess the effects of transforming growth factor alpha (TGF-alpha) on mammalian skin in vivo, we have targeted its expression to the epidermis of transgenic mice using a vector based on the human K1 (HK1) gene. Neonatal mice expressing the HK1.TGF-alpha transgene were often smaller than normal littermates and had precocious eyelid opening and wrinkled, scaly skin with diffuse alopecia. Juven...

Abstract: Background and Aim: The increasing incidence of infertility is alarming. About 10-30% of infertility are classified as Unexplained Infertility (UI) which is not an absolute clinical condition. TGF-b1 is multifunctional cytokine and produced mainly by T regulatory (Treg) lymphocytes. This cytokine plays important roles in physiology of normal pregnancy. The association of Single...
متن کاملBlocking transforming growth factor beta signaling in transgenic epidermis accelerates chemical carcinogenesis: a mechanism associated with increased angiogenesis.
Mutations in the transforming growth factor beta type II receptor (TGF-betaRII) have been identified in human cancers, which suggests a causal role for the loss of TGF-betaRII in cancer development. To directly test this in vivo, we have generated transgenic mice expressing a dominant negative TGF-betaRII (delta betaRII) in the epidermis, using a truncated mouse loricrin promoter (ML). ML.delta...
